Hi folks - My 67 year old dad has just found out that his melanoma has returned and has now metastasized on his lungs and liver. His doctor estimated he has 3 to 6 months, even though right now he looks and feels pretty healthy. We are looking at clinical trials and are impressed with what we're reading about PLX4032 and we think we can get him in to one of the trials taking place. Does anyone have any comments on the drug and the trials taking place right now?

I suggest you visit the "Melanoma International Foundation Forum"...they could be very helpful in answering this question for you. You can google it to get the web site. I Hope this helps you. |

| This compound produced very good clinical responses in Phase I trials (ASCO meeting 2009). It's reported to selectively respond to the frequently occuring B-RafV600E mutation, so it's important to check for that. It appears to be quite safe, but for a fraction of patients drug-induced formation of squamous cell carcinoma was observed (may be manageable). Best of luck for your dad. |

Thanks. This looks to be the most promising drug we've found in our research. Hope he can get in to a trial. |

My mother was diagnosed 4 weeks ago with Stage IV Melanoma that has metastasized to her lung and brain. She has gone through 15 days of direct radiation to the tumors on her brain and 5 days of temodar to treat the lung mass. I am frantically searching for a clinical trial that she qualifies for. So many of the clinical trials disqualify you if there are brain mets.
We went to MD Anderson a week ago and they advised that she has to be stable for 3 months with no new brain tumors and the existing ones cannot be any larger than when treatment started.
She had a CT Scan and MRI done this week and it showed that she is stable. We have 2 more months to go. She was admitted to the hospital this wek for dehydration and they found swelling on the brain so they put her on steroids.
Any information people can provide me is greatly appreciated.

mmlewis99 - First of all, I'm very sorry to hear about your predicament and can certainly relate. My father passed away in December. We were hoping to get him in to the University of Pennsylvania's B-RAF drug tests. He visited Penn as well as the National Cancer Institute in DC, but his health declined so fast after visiting that there was no way he could participate. There was an excellent series of articles in the NY Times last week on the test of the drug at Penn and I highly recommend you look. A good friend of mine is a dermatologist and has been following the research and we both agreed that if we could have gotten my dad in any trial, that was our first choice.

Thank you so much for your post. I am very sorry to hear about your loss. I am praying that we can find something for my mom and fast as her prognosis was gien to us as 6-8 months and that was on Feb. 1.
A friend at work gave me that article today and I started doing research on the drug. I can't tell if she would be disqualified because of the brain tumors. So many of the studies I have checked into do not treat brain tumors. She has been on Temodar and it seems to be holding them at bay for now. Did you father have brain tumors? |
